Introduction to Histological Localization

Histological localization, containing multiple technologies including high-resolution imaging, digital microscopy, and immunohistochemistry (IHC), plays important roles in clinicopathological analysis, tumor infiltration research, anatomy, and so on. Histological localization is also widely used in psychiatric disease research. For example, the visual cortex of primates can be shown, by specific staining methods, to vary locally in the pattern of lamination, and architectonic boundaries are especially obvious when the tissue is stained to reveal myelinated fibers.

Technology Applied to Histological Localization

  • IHC

IHC is an important tool in diagnostic and research laboratories. Much of the current research into the causes of neurodegenerative diseases is directed at identifying the factors that result in the formation of paired helical filaments, the deposition of beta-amyloid, cytoplasmic accumulations of alpha-synuclein, etc. Consequently, studies to localize and quantify the abnormal proteins that constitute reasons for neurodegenerative diseases are of central importance. IHC using antibodies to beta-amyloid, alpha-synuclein, ubiquitin, huntingtin, polyglutamine, and others has become a routine tool for sensitive detection and quantification of these abnormal proteins in both human tissues and in experimental animals that are used to model some of the features of these diseases.

  • Diffusion magnetic resonance imaging (dMRI)

dMRI is a specialized form of MRI that enables visualization of white matter fibers in the brain and has shown great promise as a research tool in many psychiatric conditions, including mood disorders such as depression. New advances in dMRI may provide the anatomical sensitivity and specificity to characterize the key abnormalities present in psychiatric patients at an individual level in a way previously impossible. This technological paradigm shift brings with a vastly improved capability to isolate specific tracts and measure brain networks potentially giving psychiatry a tool the equivalent of the coronary angiogram in the cardiovascular world.
